Effect of caspase inhibition on Fas-induced hepatocyte TF activity. Primary mouse hepatocytes were treated with various concentrations of IDN-7314 or vehicle (Veh, 0.1% DMSO) prior to treatment with ActD (0.2 µg/ml) and Jo2 (0.5 µg/ml). (A) Hepatocyte caspase-3 activity was measured using a NucView 488 substrate, detected using flow cytometry. The results are expressed as % positive hepatocytes gated as positive for caspase-3 activity within the total live cell population. (B) % ALT release. (C) TF procoagulant activity measured by factor Xa generation. (D) Supernatant microvesicle levels (expressed as nM PS). All measurements were determined 8 h after Jo2 treatment.
