Endocannabinoid system (ECS) modulates the function of dopamine-induced increase in cyclic AMP (cAMP) levels. E8C6 retinal mixed cell culture was subjected to cAMP quantification analysis, as described. When the selective D1 receptor agonist, SKF38393 (SKF) at 5 × 10−5 M was used, cAMP levels increased significantly. When the culture was treated with WIN after SKF stimulation, the SKF-induced increase in cAMP levels was lost. However, in cultures pretreated with AM251, a selective CB1 antagonist, cAMP levels were increased (AM251 + SKF + WIN), similar to SKF-only application. AM251 was added 15 min before SKF and WIN, which were added for 30 min, all at 37°C. ***< 0.001. N = 6 for each analysis.
