  1. U2OS wild‐type and PAWS1−/− cells were exposed to either Wnt3A or control medium for the indicated time points. Cell extracts were subjected to SDS–PAGE followed by Western blot analysis with the indicated antibodies.
  2. U2OS wild‐type (WT), PAWS1−/− (KO), PAWS1WT (+WT) and PAWS1F296A (F/A) rescue cells were exposed to either Wnt3A or control medium for 3 h followed by separation and preparation of cytoplasmic and nuclear fractions. The extracts were subjected to SDS–PAGE followed by Western blot analysis with the indicated antibodies. The lower panel represents the fold changes in active β‐catenin intensities in each fraction relative to those seen in the cytoplasmic fraction of control WT U2OS cells. The intensities of the active β‐catenin bands in each fraction were quantified by using the ImageJ software.
