Dashed purple box indicates the steps in the reaction detected by smFRET under continuous laser illumination (p_initial^obs). Solid purple box indicates p_initial^actual, which encompasses all of the events that preceed translocation in our model, including the length-sensitive k_collapse pathway that is too slow to be detected by smFRET under continuous laser illumination. For simplicity, we have shown the Nhp10 module to only inhibit k_slide (see also Discussion).
