Nup160::GFP gaps form at the nuclear rim and chromosome condensation is abnormal in lamin RNAi-depleted embryos that are depleted of dynein, related to Figures 3 and 5. Confocal fluorescence images of embryos expressing Nup160::GFP and mCherry::Histone2B. (Left)Lamin (lmn-1) RNAi-depleted and (Right)Lamin(lmn-1) and dynein (dhc-1) co-depleted embryos are shown. Sequences from sperm-derived pronuclear positioning to pseudocleavage regression are shown. Gaps in the Nup160::GFP signal appear (Arrows follow gaps in both panels) and mCherry:Histone2B appears highly condensed and localized to the nuclear rim in both conditions. The sperm-derived pronucleus migrates towards the embryo center in lamin RNAi-depleted embryos (left) but not in lamin and dynein co-depleted embryos (right). Images were acquired every 20s. Playback rate is 80X real time.
