Imaging of Halo-TERT in nuclei of living HeLa cells. 2D-movies of Halo-TERT (green, λex = 647 nm, λem = 670 nm) labeled with JF646 in the nuclei of a HeLa cells treated with no drug (Movie 1), 2 μM mismatched control oligonucleotide (Movie 2), or 2μM imetelstat (Movie 3). Movies were acquired with an exposure time of 20 ms for an effective frame rate of 46 frames per second. HA-mEOS3.2-TRF2 signals (red, λex = 561 nm, λem = 590 nm) are a maximum intensity projection of the first 50 frames of the movie acquired simultaneously to TERT. BFP-coilin signals (blue, λex = 405 nm, λem = 450 nm) are maximum intensity projections of the first 50 frames of a movie acquired immediately before simultaneous imaging TERT and TRF2 (Scale bar = 5 μm, Timestamp in ms).
