Figure 3

miR-30a negatively regulates MEF2D expression in osteosarcoma cells.

Notes: (A) Sequence alignment of miR-30a and 3′-UTR of MEF2D using mirco-RNA.org and the luciferase reporter assay. MG-63 cells were transiently cotransfected with Wt/Mut 3′-UTR with miRNAs as indicated. The effects of miR-30a on the expression of MEF2D at both mRNA (B) and protein (C) level in MG-63 cells are indicated. Data are presented as mean ± SD from three independent experiments. *P<0.05 vs control group. (D) MEF2D mRNA levels was examined by qRT-PCR in 20 cases of primary osteosarcoma tissues and adjacent nontumor tissues. (E) Correlation of miR-30a levels with MEF2D mRNA levels was examined by qRT-PCR in 20 cases of osteosarcoma tissues (Pearson’s correlation coefficient, R=−0.658). Statistically significant differences are indicated: *P<0.01 vs paired nontumorous tissues.

Abbreviations: GAPDH, glyceraldehyde 3-phosphate dehydrogenase; qRT-PCR, real-time polymerase chain reaction.
