Effect of miR-25 TuD In Vitro

(A) miR-25 TuD specificity and binding efficiency against SERCA2a 3′ UTR was assessed using a luciferase assay system. (B) Luciferase assay shows significant reduced activity by miR-25 in a dose-dependent manner in both HEK293 and H9c2. (C) miR-25 TuD transfection shows restoration of SERCA2a in H9c2 cells. (D) Recapitulation of calcium dysregulation in vitro using angiotensin II in H9c2 cells shows SERCA2a downregulation and NCX upregulation. (E) Angiotensin II upregulates pre-miR-25 and is normalized by miR-25 TuD transfer in H9c2. (F) Angiotensin II upregulates mature-miR-25 and is normalized by miR-25 TuD transfer in H9c2.
