(A) Nanobody amino-acid structure. In the H1 loop (CDR1), the Arg27 is often observed at this position in llama VHH, but is usually substituted by Tyr in dromedary VHH. This suggests that a VHH germline gene occurs in llama with the sequence GRTFSS. In framework region 2, the amino-acid sequence KQREL is often observed and is a hallmark motif for soluble, stable nanobodies. The amino-acid sequence of the H3 loop (CDR3) is given in alphabetical order. (B) SDS-PAGE gel analysis of a Nb206. M, Blue Star Prestained Protein Marker; Nb206i, sample of Nb206 after immobilized metal affinity chromatography; Nb206s, sample of Nb206 purified with size exclusion chromatography.
