Definition and processing of the 3′-end following Nei-mediated incision of mN4,5 C · G-DNA. DNA substrate (figure 1a; 1 pmol) was incubated without (lane 1) or with Nei (2.1 pmol; lanes 2–4) at 37°C for 30 min, followed by no addition (lanes 1 and 2), addition of 0.083 pmol endonuclease IV (Nfo; lane 3) or addition of 0.29 pmol T4 polynucleotide kinase (PseT; lane 4) and incubation for an additional 30 min (final volume, 10 µl).
