Cytotoxicity of fDT-bound anti-CD239 mouse mAbs on breast cancer cells. (a) Flow cytometric analysis of CD239 and HER2 expression in SKBR3 cells. As indicated in the histograms, the solid lines show anti-CD239 (left panel) and HER2 (right panel) mAbs. The grey fill indicates control IgG. SKBR3 cells highly expressed both markers. (b) Cytotoxicity of fDT-bound anti-CD239 mouse mAbs. The antibodies (87202, 87207, and BRIC221) were combined with a fragment of diphtheria toxin lacking the receptor-binding domain (fDT). SKBR3 cells were cultured in the presence of the antibodies alone or with bound fDT. Trastuzumab was used as a therapeutic antibody control. After 4 days’ culture, the number of cells was determined as described in Materials and Methods. fDT-bound antibodies suppressed proliferation of SKBR3 cells.
