Northern-blot analysis of POTLX-3 mRNA
accumulation in response to P. syringae pv phaseolicola
inoculation. cv Superior leaflets were inoculated with P.
syringae pv phaseolicola bacterial suspension (1.2 ×
108 colony forming units
mL−1) (A) or with sterile water
(mock-inoculated) (B). Plants with inoculated leaves were incubated for
0, 3, 6, 12, 18, or 24 h in the greenhouse, and total RNA was
extracted from the entire leaflet at each time. Blots of total RNA (18
μg per lane) were hybridized with the
32P-labeled 1.5-kb EcoRI fragment of
the POTLX-3 cDNA clone. Equal loading of RNA samples into
each lane was confirmed by stripping and rehybridizing the blots with a
1.2-kb wheat 18S ribosomal RNA probe.
