Sulfenylation of P450s 1A2, 2C8, 2D6, and 3A4. Prereduced enzyme was coincubated with d6-dimedone and varying concentrations of H2O2 or TCEP. Proteins were then reduced and counter alkylated with d0-iododimedone. Labeled samples were then subjected to LC-MS/MS analysis and heavy/light ratios were quantified as described in the experimental procedures. Peptides for P450 1A2 (A), 2C8 (B), 2D6 (C), and 3A4 (D) were acquired in biological duplicate and presented as means ± S.D.
