Figure 3

LDH-A promotes EMT in both SGC7901 and BGC823 cells.

Notes: (A) In SGC7901 cells, significant downregulation of ZEB2, N-cadherin and vimentin at mRNA level was detected after LDH-A knockdown, while the mRNA expression of E-cadherin was upregulated, and Snail, ZEB1 and fibronectin remained unchanged (left). Expression of ZEB2 and vimentin at the protein level was remarkably downregulated and the protein level of E-cadherin was upregulated, as shown by Western blot analysis in KD group compared with KDC group (right). (B) In BGC823 cells, significant downregulation of ZEB2, N-cadherin, vimentin and fibronectin at the mRNA level was detected after LDH-A knockdown, while the mRNA expression of E-cadherin was upregulated (left). Expression of ZEB2 and vimentin at the protein level was remarkably downregulated and the protein level of E-cadherin was upregulated, as shown by Western blot analysis in KD group compared with KDC group (right). The mRNA expression levels were normalized against β-actin. Each value represents the mean ± SD for triplicate samples. *P<0.001; **P<0.05 (Student’s t-test).

Abbreviations: LDH-A, lactate dehydrogenase A; EMT, epithelial–mesenchymal transition; KD, knockdown; KDC, KD control.
