Relationship between brain and plasma donepezil concentrations and the pharmacological effects. Donepezil HCl (0.3–10 mg/kg free drug dissolved in saline) was administered orally for four consecutive days to the corresponding treatment groups (8 animals per group). Dose-dependent changes in plasma (A) and brain (B) concentrations of donepezil were observed 1 h after the fourth dose of donepezil HCl in mice.
