TDP-43 localization in cultured neurons is XPO1 independent. (a) Hippocampal neurons were isolated and cultured. Indicated neurons were transfected with the reporter YFP-NES_PKI-NLS_SV40 and treated with either vehicle (Ethanol, 0.1% of total volume) or Leptomycin B (LMB) 10 nM for 7 hours. Nuclei were stained with Hoechst and YFP-NES_PKI-NLS_SV40 was detected with direct fluorescence. Images are representative of three independent experiments. Scale bar- 10 um. (b) Indicated neurons were treated with either vehicle (Ethanol, 0.1% of total volume) or Leptomycin B (LMB) 10 nM for 7 hours. Nuclei were stained with Hoechst and endogenous TDP-43 was detected with immunofluorescence using a TDP-43 antibody. Images are representative of three independent experiments. Scale bar- 10 um.
