Cardiac myocytes were infected with adenovirus expressing GFP-tagged HDAC5 and stimulated with 10 μM Iso and/or 50 μM PE for 1 or 24 hrs. A and C. HDAC5-GFP was immunoprecipitated with a HDAC5 antibody and detected using a phospho-specific antibody for Ser-498/661 or a HDAC5 protein antibody. B and D. Cytosolic and nuclear extracts were analyzed by western blotting with HDAC5, lamin (nuclear marker), or GAPDH (cytosolic marker) antibodies. n = 3 for each panel.
