CpG induced B10+ cells from healthy subjects increased regulatory T (Treg) cells and Tr1 differentiation from naïve T cells. Sorted B10+ cells or B10neg cells were co-cultured for 72 h with autologous CD4+CD25cells or CD4+CD45RA+CD62L+ naïve T cells stimulated with anti CD3. Treg cells, defined as CD4+CD25hiCD127lo/neg, and Tr1, defined by CD4+IL-10+, were assessed by flow cytometry. Representative flow cytometry plot (gated on CD4+ cells) and percentages of CD4+CD25 cells conversion by B10+ or B10neg into Treg cells (A,B) or Tr1 (C,D) cells are presented.
