Ovx-induced change in autophagy The hippocampus and cortex tissue collected from brains of WT-sham, WT-ovx, AD-sham, and AD-ovx groups were assessed for Beclin1, LC3, and p62 expression. Expressions of Beclin1, LC3, and p62 proteins were determined by western blot analysis. (A,D) ovx induced a significant decrease in Beclin1 expression in the 1-week group as well as a significant increase in LC3-II expression and increase in p62 expression in both the WT and 2×TgAD mice. (B,E) ovx induced a significant decrease in Beclin1 expression, increase in p62 expression, and no significant difference in LC3-II expression in the 1-month group. (C,F) ovx induced a significant decrease in Beclin1 expression, decrease in LC3-II expression and increase in p62 expression in the 3-month group. Data are presented as the mean ± SEM. Beclin1 and p62 protein expressions were relative to β-actin. LC3-II expression was relative to LC3-I. *P < 0.05 and **P < 0.01. n.s., not significant.
