Kd values for sodium and chloride
Kd values were derived by nonlinear regression analysis of Na+ and Cl− dose–response curves, obtained by measuring [3H]dopamine uptake into transiently transfected COS-7 cells during equimolar titration of 200 mm NaCl against choline chloride and sodium gluconate (final Na+ and Cl− concentrations of 200, 130, 80, 40, 20, 10, 5, and 0 mm). *, p < 0.05; ***, p < 0.001. One-way ANOVA with Dunnett's multiple comparisons test (n = 3 for variants and n = 6 for DAT WT, with triplicate determinations). Kd values for D421N could not be reliably fitted, because of the linear nature of the ion dependency curves.
