In order to quantify the expression levels of miRNA122a in hepatocytes and HCC cells, we isolated total RNA from primary human hepatocytes, HepaRG cells and HCC cell lines. qRT-PCR was then performed to determine the number of copies of miRNA122a which was reported as copy number per 1000 copies of control using the formula (2^(Ct control–Ct miRNA122a))*1000. Two-tailed t-test was performed between miRNA positive cells: HepaRG, HUCS10P, and HuH7 and miRNA negative HCC cells individually using Graph pad prism 7.0 (n > 3, ^* < 0.05, ^*** < 0.005).
