GBMX10 and GBMX16 GICs were transduced with a lentiviral vector containing a Dox-inducible shRNA against STAT3, and then transduced with wild type (WT) and mutant (Y705F and S727A) STAT3 constructs to restore STAT3 expression. (A) Cell lysates were analyzed by immunoblotting for pY705-STAT3, pS727-STAT3 and total STAT3. (B) Proliferation of control and STAT3-KD GBMX10 and GBMX16 GICs was determined CellTiter-Glo assays. (C) CellTiter-Glo based proliferation assays with the GBMX16 GICs harboring the STAT3 mutants and WT-STAT3 in the iSTAT3-KD background.
