(A) BL cells exposed to idelalisib and dexamethasone for 48 hours exhibit synergistic induction of apoptosis as measured by flow cytometry for Annexin V and propidium iodide staining in BL Ramos and Daudi cells, but no apoptosis induction in Raji or Raji 4RH cells in vitro. (B) PI3K inhibition by idelalisib leads to synergistic induction of PARP cleavage when combined with dexamethasone in Raji cells and when combined with dexamethasone or doxorubicin in Daudi cells. (C) Idelalisib exposure leads to altered expression of Bcl-2 family regulators of apoptosis. (Dex = Dexamethasone, Dox = Doxorubicin).
