Figure 1. Restricted glycolysis maintains cellular bio-energetic balance in breast cancer cells.

Figure 1

Matched pairs of either 25 mM glucose- or 10 mM fructose-adapted MCF-7, ZR-75-1, SKBR3 and MDA-MB-231 cells were seeded in 2D culture conditions and (A) glycolytic extracellular acidification rate (ECAR) and (B) mitochondrial oxygen consumption rate (OCR) were analyzed by a Seahorse BioscienceXF96 Extracellular Flux Analyzer. (A, B are n=3 to 5 from a representative of ≥ 2 independent experiments. t-test). Values expressed relative to the glucose-cultured cells. (C) Western blot for lactate dehydrogenase A (LDHA). (D) Cellular ATP abundance assay. Glucose- and fructose-adapted cells were assayed. Glucose-cultured cells were also cultured 0 mM glucose 24 h to determine the dependence of ATP concentration on glucose metabolism in these cells. (t-test). In one experiment fructose-adapted cells were also similarly transferred to 0 mM fructose (Supplementary Figure 1).
