Figure 1. ΣAVSD Value Scores the Preservation of Evolutionarily Conserved Interactions of the VSD in All VGIC Structures Deposited in the PDB.
(A) All voltage-gated ion channels (VGICs) contain a conserved structural motif within the voltage-sensing domain (VSD) as depicted by the ribbon, stick, and CPK representation of the VSD structural motif of the sodium NavAb voltage-gated channel (PDB: 3RVY), where a unique cluster of side chains forms a tripartite salt bridge network that connects amino acids from S2, S3, and S4, creating a clear ΣAVSD signature (orange line) for the VSD.
(B and C) Contact area values (ΣAVSD) of VGIC X-ray crystal structures (B) and cryo-EM-based models (C), showing the conservation of the VSD motif in all structures deposited with the more resolved structures achieving higher ΣAVSD values. The structures are identified in the graphs by their unique PDB identifying codes.
