Tespa1 deficiency selectively decreases CD40-mediated B-cell activation and proliferation. (A,B) WT (black) and Tespa1-deficient (gray) B cells were stimulated with α-CD40, LPS, or α-IgM F(ab′)2. The expression of CD21, CD23, CD44, CD40, CD25, CD69, CD80, CD86, and MHC-II was analyzed by flow cytometry on B220+ gated cells after 48 h of stimulation with the indicated molecules. (C,D) Carboxyfluorescein succinimidyl ester (CFSE)-stained B cells were stimulated with the indicated antibodies or antigen for 72 h, and proliferation was measured by flow cytometric analysis. Cells were gated on B220+. Histogram overlays depict the expression of the indicated markers. Summarized data shows the mean ± SEM from three separate experiments (n = 3), *p < 0.05.
