Validation of RNA-Seq data using qRT-PCR. Red leaf lettuce plants were exposed to different light qualities at P100 [FL, white fluorescent light; B470, blue (470 nm); G510 and G520, green (510 and 524 nm, respectively); R680, red light (680 nm)] for 24 h. Expression levels of six genes involved in flavonoid biosynthesis were subjected to qRT-PCR; they were phenylalanine ammonia-lyase (PAL), chalcone synthase (CHS), flavanone 3-hydroxylase (F3H), dihydroflavonol 4-reductase (DFR), anthocyanidin synthase (ANS) and UDP-glucose:flavonoid 3-O-glucosyltransferase (UFGT). Three biological replicates were performed for each gene. Welch’s t-test was used to calculate the p-values. Statistically significant differences in expression levels are indicated using an asterisk (p < 0.05). Gene-specific primers used for qRT-PCR are listed in Supplementary Table S3. d, day.
