Dasatinib disrupts the junctional complex of HMVEC-L and HPAEC cells in a concentration-dependent manner. HMVEC-L and HPAEC cells were exposed to dasatinib (1 nM, 10 nM, or 100 nM,) or vehicle (ctrl: 0.1% DMSO). Cells were fixed and probed with anti-VE-cadherin antibody (red), nuclei were counterstained with Hoechst 33342 (blue). (A) Representative images of two independent experiments are shown. (B) Quantification of the immunofluorescence staining intensity. Arrows indicate loss of junctional protein staining, while arrowheads indicate the gaps between cells. Data analysis was done by one-way ANOVA with Tukey post hoc test. Values are presented as mean ± SD; p < 0.05, +p < 0.001, p < 0.0001 compared to ctrl (N = 6).
