Altered expression of ECM proteins in ChECs. A: PEDF^+/+ and PEDF^−/− ChECs were plated on gelatin-coated 60-mm dishes and incubated for 48 h in serum-free growth medium. The conditioned medium and cell lysates were collected for analysis of ECM proteins by Western blot analysis as described in materials and methods. The expression of TSP1, PEDF, TSP2, fibronectin, osteopontin, SPARC, and collagen IV was determined using specific antibodies. Note the increased expression of osteopontin in PEDF^−/− ChECs compared with PEDF^+/+ cells. β-Actin was used as a loading control for cell lysates (bottom).
