The effects of BET inhibitor on PDAC cells. (A) Panc-1, BxPC-3, and HS766T cells were treated with increasing does of JQ-1 or I-BET762 for 72 h. Cell viability was determined by the CCK-8 assay. (B) Indicated cell lines were treated with 1 μM JQ-1 or 1 μM I-BET762. DNA synthesis was analyzed by the Cell-light EdU Apollo 488 in vitro assay. (C) Panc-1, BxPC-3, and HS766T cells were treated with 1 μM JQ-1 or 1 μM I-BET762 for 24 h. The percentage of cells in different phases of cell cycle was analyzed by flow cytometry. (D) PDAC cells were treated with 1 μM JQ-1 or 1 μM I-BET762 for 24 h. Early and late apoptotic cells were analyzed by flow cytometry. (E) PDAC cells were treated with the 1 μM JQ-1 or 1 μM I-BET762for 24 h. The indicated protein levels were analyzed by western blotting.
