Effect of pH on PLD activity. The optimal activity of PLD was determined using standard assay in 40 mM of the following buffers. Symbols: diamond, disodium hydrogen phosphate-citric acid buffer (pH 4.0–8.0); square, Tris-HCl buffer (pH 8.0–9.0); triangle, glycine-sodium hydroxide (pH 9.0–10.0). The maximum activity was taken as 100%. To determine pH stability, the enzyme was incubated at 37°C for 60 min in various buffers mentioned above, and the residual activities were measured.
