Ratio of intact, permethylated glycan yields produced by the SCF procedure relative to a commonly employed SC procedure (SC 50 min):²⁰ (a) O-glycans from Fetuin and (b) N-glycans from bovine ribonuclease B. For each glycoform, the highly-averaged intensities of the two MALDI-MS peaks corresponding to the two permethylation procedures conducted with light and heavy labeling reagents were compared and presented here as the SCF/SC ratio. Two-tailed Student’s t-test comparing the SCF/SC ratios to the theoretical value of 1 demonstrated significant differences between the two procedures as indicated (p < 0.05).
