Intracellular insulin staining reliably detects β cells for further phenotypic analysis by flow cytometry. Shown is the gating strategy for pancreatic islets harvested from a 5 week NOD mouse. β cells were identified as forward and side scatter high cells, CD45.1⁻ cells, CD4⁻ and lineage negative (CD8⁻, CD11c⁻, CD11b⁻, B220⁻, F4/80⁻), live, intracellular insulin-positive cells.
