Blockade of CD70 reduces primary expansion of CD8 T cells but not antibody responses after modified vaccinia virus Ankara (MVA) immunization. C57BL/6 mice were immunized intravenously with 5 × 107 TCID 50 recombinant rMVA (rMVA) alone or in combination with 300 μg anti‐CD70 (FR70) intraperitoneally on days 0, 4 and 7. Peripheral blood lymphocytes were gated on CD4 CD8+ CD44+ B820‐27‐dextramer+ T cells and further analysed for CD62L and CD127 expression (a). The frequency of B8‐specific CD8 T cells over time is shown in (b). CD127 expression on B820‐27‐specific CD8 T cells is shown as geometric mean fluorescence intensity (GMFI) (c). The total number (d) and memory phenotype (e) of splenic B820‐27‐specific CD8 T cells was determined on day 84. MVA‐specific IgG titres in the serum on day 21 were determined by ELISA (f).
