Spt20 is degraded in the presence of tra1 $_{Q3}$ . Strains CY7530 (TRA1) or CY7531 (tra1 $_{Q3}$ ) containing TAP-ADA2 and YCplac33-SPT20-myc⁹ were grown to saturation in media lacking uracil, diluted 1:10 in YPD and grown for eight hours. Cells were harvested and lysed using glass beads. 200 μg protein was separated by SDS-PAGE (10%), transferred to a PVDF membrane and blotted with an anti-myc antibody.
