Spt7^SLIK increases due to stress caused by tra1 $_{Q3}$ . Diploid strains CY7405 (Flag⁵-TRA1/TRA1), CY7406 (Flag⁵-tra1 $_{Q3}$ /TRA1) and CY7407 (TRA1/TRA1) containing myc⁹-SPT7 on a URA3 centromeric plasmid were grown to an OD $_{600}$ ∼2.0 and protein harvested by grinding in liquid nitrogen. Tra1 was pulled down using anti-Flag M2 magnetic beads from 2 mg of crude lysate. Protein was separated by SDS-PAGE (5%). Spt7 and Tra1 were detected with anti-myc and anti-Flag antibodies respectively.
