p66Shc and MAPK activity in M238/M238R after combined PEITC and inhibitor treatment. Cells were seeded in six‐well plates at a density of 500 000 cells per well and treated with 20 μm of AZD6244, PLX4032, or SP600125, 1 hour prior to treatment with 20 μm PEITC for 30 min. Immunoblots were performed using phosphorylation‐specific and total protein antibodies for p66Shc (A) and JNK1 (B). (C) For ROS measurements, cells were seeded in 8‐well chamber slides and treated as described above before staining with 100 μm MitoTracker Red CM‐H2XRos for 10 min.
