A molecular bellows mechanism for the MacAB-TolC pump driven by mechanotransmission. The proposed catalytic cycle of the MacAB-TolC efflux pump (Top) is shown in comparison with the operation of a fireplace bellows (Bottom). Substrates are driven out by mechanotransmission-induced changes in the MacB periplasmic domains and prevented from flowing back into the periplasm by a valve in MacA.
