Two-step model for assembly of new dCENP-A at centromeres. (A) Table summarizing the data concerning the centromeric localization of proteins tested in Figs. 3 and 6. (B) Putative model for the assembly of new dCENP-A at centromere: (1) CAL1-targeted new dCENP-A/H4 associates with centromeres, but incorporation is blocked by previously incorporated placeholder nucleosomes; and (2) FACT-mediated transcription induces the eviction of nucleosomes, thus allowing the full incorporation of new dCENP-A.
