Detection of Luciferase Activity after Transfection of HEK923 Cells with Synthetic hGLuc mRNA
3 × 105 HEK293 cells were incubated for 4 hr at 37°C with 0.2, 0.5, or 1.5 μg mRNA complexed with Lipofectamine 2000. Then transfection complexes were discarded, and fresh medium was added to the cells. After 24 hr, the luciferase activity (RLUs) was detected in the supernatants and cell lysates. Cells treated with medium or medium and Lipofectamine 2000 (L2000) served as negative controls. Results are shown as mean ± SEM (n = 3). Statistical differences were determined using two-way ANOVA followed by Bonferroni’s multiple comparisons test (**p < 0.01, ***p < 0.001, ****p < 0.0001).
