(a) PLA was performed for detection of Mincle-CD11b and Mincle-Syk interactions in iBMMs that were stimulated with TDM for 12 h. (b) The Mincle–CD11b interactions following stimulation with TDM (50 μg ml⁻¹) for 12 h or LPS (100 ng ml⁻¹) for 6 h were compared. (c) Interactions of CD11b/CD18 with endogenous talin in WT and CD11b^−/− iBMM cells were examined after stimulation with TDM (50 μg ml⁻¹) for 24 h or fibrinogen (1 μg ml⁻¹) for 30 min. Interactions were visualized as fluorescent spots (red, PLA signal), and the nuclei were stained with DAPI (blue). The number of PLA signals was determined for at least 50 cells for each condition.
