EBV B95-8 bacmid 2089 infected 293 cells were transfected with Zta expression vector (+Z) to induce EBV lytic replication and fixed at 24hr (24h + Z), 48 hr (48h + Z) or 96 hr (96h + Z) after transfection. Fixed cells were stained for RNA Pol II (Red) and BMRF1 (Green) to visualize the formation of replication compartments. Cells were also treated with PAA to block viral replication at 24 hr (24h + PAA) or 48 hr (48h +PAA) post-induction and fixed at 96 hr post-transfection to assess the effect of blocking DNA replication on replication compartment structure. Enlarged images of replication factories are shown in each panel and arrows indicate magnified cell. BMRF1, the polymerase-associated processivity factor was used as a marker for viral replication foci.
