Manual cell counting was performed to quantify the morphological changes of viral replication factories. 15–20 fields of slides were randomly chosen for counting under the objective magnification of 63X. Cell numbers were expressed as percentage of replicating cells (BMRF1 positive cells). Each cell was categorized as either demonstrating colocalization if greater than 75% of the BMRF1 and BGLF4 signal was overlapping. (A) Percentage changes in co-localization between BMRF1 and BGLF4 after PAA treatment at either 24 or 48 hr post-induction were calculated. * p value < 0.0001, ** p value <0.001.
