PSCA antibody-targeted MSNs deliver function miRNA cargo to prostate cancer cells. PEGylated MSNs covalently linked to an antibody against prostate stem cell antigen (PSCA) were loaded with either Dy547 labeled miRIDIAN microRNA hairpin inhibitor or mimic negative controls or miRIDIAN microRNA mimic against miR-221-3p (Dharmacon). (a) Epifluorescent (grayscale) and DIC images show uptake of Dy547-labed negative control molecules by PC3 cells. These nucleic acid molecules appear as dots in the grayscale image and are present in nearly all of the cells. (b and c) RNA was isolated 72 hr after treatment with MSNs loaded with mimic negative control or miR-221-3p mimic and qPCR levels of (b) miR-221-3p or (c) the validated target HECTD2.
