Human first trimester cytotrophoblast cells form spheroids which contain proliferative cells and express both ES and TS cell markers. (a) Shows spheroids formed on days 4 and 8 by freshly isolated p-trophoblast and co-immunostaining for CK7 (trophoblast marker) and Ki67 (proliferation marker) in the first generation. Cells dispersed from the first generation spheroids also produced spheroids during the second generation and stained positive for CK7 and Ki67 (b). (c) Shows co-expression of ES markers OCT4, NANOG in cells within the p-trophoblast spheroids. (d) Shows co-expression of TS makers CDX2, ELF5 in cells within p-trophoblast spheroids.
