Figure 1.

Figure 1

Thymus phenotype and function. (A) Representative flow cytometry plots showing the four thymic subpopulations double-negative (DN) (CD4CD8), double-positive (DP) (CD4+CD8+), CD4+ single-positive (SP) (CD4+CD8), and CD8+ SP (CD4CD8+) cells, identified within the CD90.2+ population. FoxP3+ Treg cells are studied inside the CD4+ SP population. 30,000 events are recorded. (B,C) Frequencies of the four thymic T-cell subpopulations (B) and FoxP3+ Treg cells (C) in thymus of KO (white circles, n = 12–16) and wild-type (WT) (black circles, n = 5–9) mice at 3, 6, or 18 months. Data (mean ± SEM) are representative of two to three independent experiments. Unpaired t-test was used for statistical analysis. (D) sjTREC, dβTREC, and sj/dβ ratio in splenocytes of KO (white circles, n = 12) and WT (black circles, n = 6–12) mice at 3 and 6 months are shown. Data (mean ± SEM) are representative of two independent experiments. Unpaired t-test was used for statistical analysis. ***p < 0.001, **p < 0.01, *p < 0.05.
