SUN1 inhibition of HIV-1 infection is dependent on the nuclear import process. (A) Dimethyl sulfoxide (DMSO) or aphidicolin A was added to the culture medium of HEK293A cells 2 h before infection. The cells were infected with VSV_G-pseudotyped wild-type (WT) NL4-3luc or its mutants containing the indicated CA mutations. At 48 h postinfection, luciferase activity was measured. (B) HEK293A cells transduced with an empty lentivector (EV) or a lentivector expressing SUN1-Flag were infected with the indicated VSV_G-pseudotyped viral vectors. At 48 h postinfection, luciferase activity was measured. Fold inhibition (FI) was calculated as the luciferase activity in the control cells divided by that in the SUN1-expressing cells and is indicated above the bars. Data presented are means and SD for three independent experiments.
