Whole-cell fractionation of HaCaT, THP-1 and HGT-1 cells shows that VPS33B and VIPAR form a complex that is considerably smaller than CORVET/HOPS and does not contain CCDC22. THP-1 (A), HaCaT (B) and HGT-1 (C) cell lysates were injected onto a Superose 6 10/300 GL gel filtration column and eluted fractions were analyzed by immunoblotting using the antibodies indicated. Additional bands at unexpected sizes in panel C, which are presumed to arise due to cross-reactivity of the polyclonal antibodies, are marked with open arrowheads.
