Figure 2

MALAT1 knockdown inhibited proliferation and migration and promoted apoptosis in PC cells.

Notes: DU145 and PC3 cells were transfected with si-MALAT1 or its scramble control (si-NC). (A) At 48 h after transfection, MALAT1 expression was detected using RT-qPCR assay. (B) Two weeks post transfection, colony formation analysis was performed. (C) At the indicated time points (0, 24, 48, and 72 h) following transfection, cell proliferation patterns were determined by MTS assay. (D) At 24 h after transfection, cell migration capacity was examined by transwell migration assay. (E) At 48 h after transfection, apoptotic rate was determined using flow cytometry via PI and Annexin V-FITC double staining. (F) At 48 h upon transfection, protein expressions of cleaved PARP, cleaved caspase-3, BAX, and Bcl-2 were measured by western blot assay. *P<0.05.

Abbreviations: MALAT1, metastasis-associated lung adenocarcinoma transcript 1; PARP, poly (ADP-ribose) polymerase; PC, prostate cancer; PI, propidium iodide; RT-qPCR, reverse transcription-quantitative polymerase chain reaction.
