Table 4.

Number of models exhibiting fluctuations in diastolic Ca2+ in the Grandi-based in silico population calibrated with AP + ICaL + IK1 data in the absence and presence of interventions on Jrel and INCX.

Following AP + ICaL + IK1 Calibration 90% Block Jrel 100% Increase Jrel Increase INCX to Normal Levels
Number of models with spontaneous Ca2+ release 4 1 4; more peaks and smaller amplitude of oscillations 0

AP, action potential; ICaL, L-type Ca2+ current; IK1, inward rectifier K+ current; Jrel, release current; INCX, Na+/Ca2+ exchanger current. 90% block of Jrel abolishes fluctuations in 3 of 4 models; 100% increase in Jrel exacerbates diastolic Ca2+ fluctuations (more peaks in diastolic Ca2+, smaller amplitude of Ca2+ fluctuations). Increasing INCX to normal levels abolishes fluctuations in all models.
